Los Santos: un'enorme e soleggiata metropoli piena di sedicenti guru, attricette e celebrità sul viale del tramonto. Un tempo era l'invidia del mondo occidentale, ma ora è costretta ad arrangiarsi per restare a galla in un'epoca di incertezza economica e TV via cavo da quattro soldi. In mezzo a tutto questo, tre criminali molto diversi tra loro si danno da fare per sopravvivere e realizzarsi, ognuno a modo suo: Franklin, un ex gangster di strada, è a caccia di soldi e di opportunità; Michael, un ex criminale professionista, sta facendo i conti con una "pensione" meno rosea del previsto; Trevor, un maniaco violento, pensa solo a farsi e al prossimo, grande colpo. I tre non hanno altra scelta: decidono di rischiare il tutto per tutto in una serie di colpi audaci e pericolosi che potrebbero sistemarli per la vita.

Il gioco sarà ambientato nello stato di San Andreas (ispirato alla California e al Nevada), nella città di Los Santos (ispirata alla Los Angeles) e nella riserva naturale di Blaine County (ispirata alla Contea di Blaine in Idaho), la campagna e il deserto attorno. Rispetto a quella presente su Grand Theft Auto: San Andreas, la città è stata completamente ricreata e resa molto più grande e più fedele alla sua controparte reale. Si tratta di una mappa molto vasta che comprende spiagge, coste, vigneti, campagne, aree selvagge, la zona del Monte Chiliad (e altri monti) completamente esplorabili, la regione di Salton Sea, una base militare e ovviamente la zona suburbana e il centro cittadino di Los Santos. Sarà anche possibile esplorare l'ambiente subacqueo e grande importanza è data allo spazio anche in verticale, con diversi piani esplorabili in altezza. L'anteprima mondiale di Game Informer ha rilevato che l'ambientazione del gioco è più grande di quella di Red Dead Redemption, Grand Theft Auto IV e San Andreas messi assieme; dalle prime informazioni trapela che il territorio di GTA V possa arrivare a una dimensione di oltre 200 Km quadrati.

Personaggi:

• Franklin Clinton: venticinquenne afro-americano, è il primo protagonista disponibile per l'uso, è impiegato in una concessionaria di auto di lusso armena, che vende le auto a chi non se le può permettere, e quando non riescono a pagare, lui ha il compito di recuperare le auto con gli interessi.
• Michael De Santa (pseudonimo di Michael Townley): un rapinatore di banche "in pensione", sui 40 anni, che vive sotto il programma di protezioni testimoni del FIB (la controparte dell'FBI), ha una moglie, Amanda, che spende tutti i suoi risparmi, e due figli, Tracey e Jimmy con cui non ha una comunicazione molto attiva.
• Trevor Phillips: amico di Michael ed ex veterano di guerra con esperienze di volo, vive in una roulotte nella località desertica di Blaine County, ha un carattere maniacale e assolutamente psicopatico. Gestisce la "Trevor Philips Enterprises", società che si occupa di produrre e spacciare droga in giro per la contea.
• Lamar: migliore amico di Franklin, è un ragazzo lunatico e divertente, appartenente a una gang dai colori verdi in conflitto con la gang dei Ballas.
• Jimmy: è il figlio di Michael, ventenne pigro e fumatore di marijuana.
• Tracey: figlia di Michael, vuole diventare una pornostar.
• Amanda: moglie di Michael, ha una personalità forte e litiga spesso con Michael.
• Dr. Friedlander: è il terapista di Michael.
• Simeon Yetarian: di origini armene, è il proprietario del concessionario in cui lavora Franklin. Verrà colpito più volte da Michael.
• Ron: migliore amico di Trevor, è un complottista teorico, totalmente paranoico.

Visitors to the greater metropolis of Los Santos and the countryside of Blaine County will encounter faded celebrities, party people, hikers, bikers, and every other manner of colorful denizen. You'll be able to traverse everywhere from the tops of the mountains, through the streets of Los Santos and to the depths of the ocean floor.
• Michael, Franklin and Trevor - Three playable protagonists - Michael, Franklin, and Trevor may live in different parts of town, have their own issues, attitudes, lifestyles, desires and goals - but they do know how to work together. They also each have their own unique skillsets to bring to the table when it's time to get the job done.
• Heists - Petty crimes, hustling, and stealing cars can only get you so far in Los Santos. To score big, Michael, Franklin and Trevor will work together to pull off grand Heists - big, multi-part missions that require careful preparation, recruiting, and precise (and often explosive) execution. Cash is king in GTAV and while there will be plenty of ways to acquire and spend it - heists are the way to earn the big bucks.
• 'Recreational' Activity - The open world of Grand Theft Auto V is not just massive in scale, but will offer more to get into than ever before. For recreation, play a bit of golf or tennis, zen out with some yoga or cycle through the countryside. For adrenaline heads, parachute over the city, or customize your Cheetah and take it to the streets to race suckers for money. Or earn cash the old fashioned way, by taking it - whether by ripping off liquor stores or by buying and taking over businesses that will earn you dividends. Even minding your own business having a stroll you may meet one of Los Santos' weirdos or starlets and find yourself off on a wild misadventure you never expected.
